Résumé : | In this work, we answered the question, did IgA play a role in pulmonary inflammation and fibrosis. To achieve this goal, we compared the lung responses to silica particles between wild-type mice (WT) and IgA-deficient mice (KO). Control group of mice were exposed to saline solution. Treated groups corresponded to silica instillation in WT and KO animals. We determined the acute inflammation (3 days), chronic inflammation (4 months) and chronic fibrosis (4 months). We analysed lung inflammation by measuring levels of inflammatory mediators (CCL2 and KC) and by evaluating pulmonary leukocyte accumulation (neutrophils, macrophages or lymphocytes). Our data indicated that acute and chronic inflammation was comparable between WT and KO treated mice. Similarly, biomarkers of lung fibrosis development (TGF-B, TIMP-1, as well as collagen) were not modified in silica-treated mice. These last data were confirmed by histological analyses. In conclusion, our results suggest that IgA has no major role in the development of silica-induced acute or chronic inflammation or fibrosis. However, further studies are needed to consolidate our knowledge, as not all questions have yet been answered. |
