Résumé : | Peroxiredoxins (Prxs) are highly present proteins in every cell and possess three functions. The first is hydrogen peroxide (H2O2) detoxification, which makes these proteins antioxidants since H2O2 is known to be a reactive oxygen species (ROS) that causes oxidative stress. This phenomenon is responsible of the ageing of cells which can lead to diseases linked to age such as Alzheimers and Parkinsons. However, it was recently discovered that H2O2 also regulates many physiological processes and that Prxs could function as signal mediators and chaperones. Indeed, a certain category of Prxs, the typical 2-Cys Prxs, are capable of oligomerizing into dimers and decamers to function as chaperones. This class comprises two structurally very similar isoforms with slightly different functions. It has always been considered that the two isoforms assemble independently of each other in homo-oligomers but a possible hetero-oligomerization between them has barely been explored. It is surprising because this could be at the origin of oligomers with new properties that could improve the detoxification of H2O2 or alter its use as a signal molecule. In this work, we aimed to investigate the hetero-oligomerization of typical 2-Cys Prxs from different eukaryotes i.e., plants and fungi. For this, we used tsa1/2 yeast cells as reaction compartments to co-express two genes encoding Prxs isoforms, one tagged with roGFP2 to induce size-shifts between homo- and hetero-oligomers usually identical in size but also between the hetero-decamers depending on the isoforms ratio they have to be able to separate them on CN-PAGE by different migration distances. |
