Multiple myeloma is the second most common hematologic malignancy. Therefore, there is an increasing clinical interest in the measure and achievement of minimal residual disease (MRD) negativity in the bone marrow of MM patients, as defined by Multicolor Flow Cytometry (MFC).
For this study focused on the research of the MRD of MM, we used two tubes of analysis: a surface tube and an intracytoplasmic one. The surface and intracytoplasmic panels have a specificity that one highlights the expression of markers CD81 and CD200 and the other allows to stand monoclonality kappa or lambda respectively. The CD38 ME method or surface tube is used for the patients treated with daratumumab. We evaluated several parameters for the validation of the MRD of the MM: linearity test, carry-over and limits of blank, detection and quantification (LoB, LoD and LoQ). For both techniques, we obtained consistent and satisfactory results. However, we cannot only conclude based on five marrow samples that both methods can be validated for research of MRD of MM, more samples would be needed to reach this conclusion. Nevertheless, we can already say that the surface method used is more sensitive than intracytoplasmic method according to the results obtained, this could be confirmed during the next studies. |
