Inflammation is a phenomenon that involves a large number of cellular and chemical actors. Indeed, as well as numerous immune cells, several molecules intervene in order to eliminate the stimulus following an injury. These mediators involved in inflammation can be either pro- or anti-inflammatory. During inflammation, a sensation of pain is often
perceived. This sensation of pain is due to activation of the nociceptive system by inflammatory mediators. However, after an operation, the acute pain due to tissue injury may persist and never fully resolve leading to chronic inflammation and pain. Among the mediators involved in inflammation, we are particularly interested in bioactive lipids such as endocannabinoids. The endocannabinoids are endogenous mediators that bind the CB1 and CB2 cannabinoid receptors. Once activated, these ubiquitous receptors have many beneficial effects, including the reduction of pain. The endocannabinoid of interest in this study is 2-arachidonoylglycerol (2-AG). It is produced from membrane phospholipids and hydrolyzed by specific enzymes, such as monoacylglycerol lipase (MAGL). To assess the effect of increasing the endogenous levels of 2-AG on post-operative pain, we used the hind paw incision (HPI) model in mice and we administered JZL184, an inhibitor of MAGL). First a measurement of mechanical hyperalgesia was performed on control mice and operated mice that were treated or not with JZL184. These measurements showed that mice treated with JZL184 had a higher pain threshold than untreated mice. Secondly a measurement of the expression of mediators of inflammation TNFa and MCP-1 was made to see if there was a difference between the operated mice and the control mice. In conclusion, further results will need to be obtained to demonstrate the true effectiveness of JZL184 by showing a decrease in inflammation. |
