Mice produce ultrasonic vocalizations (USVs) in a variety of social situations. USVs are ultrasound, waves of very high frequency (more than 20 kHz) and inaudible by the human ear. Mice use them to communicate with their own kind. These USVs are useful to study many neurological diseases including autism and schizophrenia.
A previous study showed that pups produce isolation calls, a common USV, when theyre isolated from their mother for a certain time. They also produce USVs when their body heat drops. Adult male mice also have this capacity during exploratory behavior and in sexual situations, when theyre exposed to fresh female urine, anesthetized female or not. Therefore, the goal of my work is to develop a system for measuring ultrasonic vocalization in mice. To do so, I set up a system containing a recording box, a test cage, a microphone linked to a computer with the recording software UltraVox XT. I used several groups of C57BL/6 mouse model for a total of 46 mice. I put couples together during 3 weeks to have a lot of pups. From 4 or 5 days after birth, I isolated them from their parents before the beginning of the test. Concerning adult males, a first group was tested with some sexual stimuli and a second group was tested during exploratory behavior in a new environment with a dim light. My work shows that there is a difference between the number of USVs on the fourth day after birth (which are less) and the other days. But no difference in duration of vocalizations. We noticed that the pups can emit 3 different frequency ranges (51-67 kHz, 80-99 kHz, 118-142 kHz) and the lowest frequencies are the most numerous. There is a significant difference between the duration of the USVs and the frequency. Unfortunately, I have no potent results in adult mice. In conclusion, we show that our measurement system works. The UCLs Cell Physiology laboratory will be able to use this test and compare it to other pre-existing social interaction tests. |
